L’acide vaccénique ([AV], un acide gras trans 11‑18:1), l’AGT prédominant produit par les ruminants, est un précurseur de l’acide linoléique conjugué qui peut améliorer l’état des patients atteints de dyslipidémie athérogénique et atténuer les symptômes de stéatose hépatiques dans les modèles animaux. Notre étude visait, d’une part, à évaluer la biodisponibilité intestinale de différentes sources d’AV, dont les triglycérides naturels des ruminants et les acides gras libres (AGL) synthétiques, et, d’autre part, à déterminer les voies mécanistiques qui modulent la bioactivité de l’AV. Méthode et résultats. In vitro, l’AV agit comme un agoniste partiel des récepteurs α et γ d’activation et de prolifération des peroxysomes (PPAR), avec une affinité semblable à celle des agonistes connus des PPAR. Nous avons également confirmé qu’à des concentrations de 30 et de 100 μM, l’AV élimine l’hypertrophie des cardiomyocytes in vitro, et ce, de façon proportionnelle à la dose d’α et de γ‑PPAR administrée. L’administration in vivo d’AV (1 %, p/p) a donné lieu à une augmentation du nombre de transcrits d’ARNm et de l’expression des γ‑PPAR dans la muqueuse des rats JCR:LA-cp, un modèle du syndrome métabolique (p < 0,01 et p < 0,05, respectivement). De plus, la biodisponibilité intestinale in vivo de l’AV issu de triglycérides était supérieure à celle de l’AV provenant d’AGL (p < 0,01). Conclusion. L’activation des voies dépendantes de l’α‑PPAR et du γ‑PPAR explique, en termes mécanistiques, comment l’AV peut améliorer les lipides du sang et influer sur les troubles métaboliques associés à ces lipides dans les cas d’hyperlipidémie. Nos résultats viennent également appuyer la déclaration différentielle, sur l’étiquette, des AGT produits lors de la transformation par rapport à ceux de source naturelle.
